Problem: The acquired image is not in focus
Possible cause
Remedy
The microscope was not focused
on the sample.
Focus the microscope on the sample.
The parfocality between the
camera adapter and the
eyepieces is not adjusted
properly.
Adjust the parfocality between the
camera adapter and the eyepieces
properly.
The aperture diaphragm of the
condenser is too wide open.
Set the aperture diaphragm to 75% of the
objective’s numerical aperture and focus
the microscope on the sample.
The field diaphragm is too wide
open.
Narrow the field aperture appropriately
and focus the microscope on the sample.
Lens components of the
microscope and the cover glass
on the bottom of the camera are
dirty.
Clean the objective, condenser, window
lens of the microscope, and the cover
glass on the bottom of the camera (see
page 21).
The microscope and camera are
being subjected to vibration
during acquisition.
Acquire images in an environment where
the microscope and camera are not
subjected to vibration. It is effective to use
an anti-vibration table.
