Press the BLOOD IN button and open the clamp on the blue-striped tubing.
Valve V1 opens, allowing processing/wash solution to flow by gravity from the
container connected to the blue-striped tubing to the cell product bag.
Manually agitate the cell product bag to ensure adequate mixing (Figure 6-18
on page 6-20).

After the desired volume of processing/wash solution has entered the cell
product bag, press the STOP RESET button to close valve V1.

Close the slide clamp on the blue-striped tubing.

If additional predilution with another processing/wash solution is desired,
proceed to step 9. If no additional predilution is necessary, proceed to step 11.

If you want to use the fluid connected through valve V2 for a second
predilution, press the PREDILUTE button. Valves V1 and V2 open, allowing
the second processing/wash solution to flow by gravity into the cell product
bag. Manually agitate the cells as the processing/wash solution flows into the
cell product bag.

After the starting cell product has been prediluted with the desired volume,
press the STOP RESET button to close valves V1 and V2.

Allow the mixture to equilibrate for the appropriate time.

Hang the prediluted cell product bag on the left hanger bar.

Remove the clamp from the tubing between the rotating seal and the RCD.

Press the BLOOD IN button. Valve V1 opens, allowing the starting cell
product to flow by gravity into the cell processing bag.
After a short time, the starting cell product stops flowing. Depending on the
initial volume, some starting cell product may be left in the cell product bag.

Set the CENTRIFUGE SPEED control to 3000 rpm, then press the AIR
OUT button. Valve V1 remains open and the centrifuge spins, pushing air and
starting cell product back up the red-striped tubing through valve V1 to the
cell product bag.

When air and starting cell product reach the cell product bag, press the
BLOOD IN button again. Additional starting cell product enters the cell
processing bag.

When no more fluid will flow into the cell processing bag or the fluid level
(Figure 6-15 on page 6-17) is just above the rotating seal, press the STOP
RESET button.
The capacity of the cell processing bag is approximately 630 mL. If the volume
of the starting cell product is more than 630 mL, not all of the starting cell
product will flow into the cell processing bag during the Blood-In/Air-Out
sequence. Additional starting cell product may be added during subsequent
Agitate/Wash-In operations by programming valve V1 to open instead of
valves V2 or V3. Add additional wash cycles as needed.
