7.3 End of collection
At the end of the sampling period a signal tone sounds and the message “End of sampling” is displayed.
Press Enter to stop the signal and go back to the SAMPLE menu.
Remove the impaction grid and close the Petri dish. Place the Petri dish into an incubator.
In case of problems occurring during the cycle, the error codes are successively displayed at the end of the
cycle. Press ENTER to acknowledge each Error message. Some errors may stop. In this case, the message
“Sample interrupted” is displayed.
7.4 Achieving quality sampling
To get good quality sampling a standard methodology is required. Indeed large ranges of factors have an
impact on the harvest of micro organisms and their growth (air movements, atmospheric pressure, impaction
speed, physiological state of the micro organisms).
Quality medium sampling
The users are free to choose the media which are the most relevant to grow the organisms they wish to isolate.
In any case, they must ensure that the following precautions are taken:
The depth of the agar in the Petri dish should be reproducible and measure from 2,5 to 3,5 mm
The agar surface should be flat and levelled.
The storage methods should be appropriate to the medium
The agar should not present any dryness
The agar should not present water droplets when used. Any excessive humidity on the agar may affect
results
Some adapted media, suggested by BioMérieux S.A suit perfectly this kind of analysis.
7.5 Collection Method
Different procedures need to be followed in order to prevent fluctuations due to exterior phenomena or users.
Those procedures should be strictly followed when collecting in Zones 3 and 4.(see § 7.7.1).
7.5.1 Precautions for a good collection
- Hands and nails should be washed before using the Sampl'air Lite (using products such as PROSEPTOL
®
is
recommended)
- Protection clothing should be worn in accordance with the collection environment.
- Avoid breathing and coughing in direction of the Petri dish when open.
- Complete the work quickly and efficiently and avoid any delays.
7.5.2 Collection procedures
- Collect from low contaminated areas moving up to higher contaminated areas.
- It is recommended to achieve sampling in the middle of the room or in different points especially when the
size of the room is significant.
- Duplicate sampling (at the same place) in order to eliminate false results and to allow a rate of contamination
calculation.
If the instructions above are not followed, the measurement parameters
might be difficult to reproduce and the results might be false. Besides, some
media, which have been developed for clinical bacteria, are not
recommended for environment microorganisms research.
