Instrument Description and Main Features
The following components are necessary for operating the Laser TIRF imaging system in combination with
Axio Observer:
−
Microscope Axio Observer.Z1 (431007-9901-000 and higher) with basic laser safety kit, version 2,
−
Laser safety device for microscope and peripheral units,
−
Laser Bench Tower with up to 2 lasers or Laser Bench Rack with a maximum of six lasers,
−
Slider Laser TIRF with monomode fiber (pre-adjusted and coupled to Slider Laser TIRF),
−
Objective as per list with NA greater than 1.45,
−
Control PC with at least 4096 MB working memory,
−
Monitor for image display,
−
Digital camera with more than 10 bits, e.g. AxioCam MRm, for acquisition and documentation.

ZEN software (blue edition),
−
Vibration-damped table or anti-vibration plate for Axio Observer,
The Laser TIRF imaging system is optimized for polarization-retaining monomode fibers. When
using fibers from other manufacturers, please note the instructions given in Section 2.14 on
page 51.

Slider Laser TIRF, man. / mot.
The Slider Laser TIRF is inserted into the receptacle of the luminous-field diaphragm integrated in the
microscope. The optical structure of the Laser TIRF imaging system allows the use of conventional epi-
fluorescence illumination and TIRF illumination sources without any modification or removal of the Slider
Laser TIRF. The specimen can be illuminated either alternately in rapid succession or simultaneously with
both light sources.
The Slider Laser TIRF is available in a manual and a motorized version (Fig. 2-2, see next page). In the
Slider Laser TIRF mot. the TIRF angle is set by means of a software-controlled motor; in the Slider Laser
TIRF man. it is set by means of an adjusting screw.
