2. Auxiliary Phases
Substrate Sampling
User Action
System Auto Sequence
User initiates Substrate Sampling at the HMI.
Phase window pops up for the user to enter the following setpoints:
• Air MFC Flow Rate (SCCM)
• Agitator Speed (rpm)
• Stabilization Delay Duration (min.)
• Recirculation Pump Ramp Duration
User enters setpoints, and selects the play
button.
Records loop outputs for temperature, pH, and DO equipment
modules.
Stops automated control of all equipment modules and pumps.
Sets the Air MFC Flow Rate to the Substrate Sampling Air Flow Rate
and Agitator at the Substrate Sampling Agitator Speed.
Turns all heaters off, except the exhaust heater.
Stops all pumps.
Sets recirculation pump to counterclockwise in Manual jog mode.
Prompts user to manually jog recirculation pump to desired media
volume in the FBR.
User manually jogs recirculation pump to
desired media volume in the FBR and confirms
prompt at the HMI.
Prompts user to manually disconnect the FBR for sampling in a sterile
cell culture hood.
User conducts substrate sampling in a
biosafety cabinet. When complete, reconnects
the FBR.
Prompts user to confirm when the FBR and FBR Temperature sensor is
reconnected to resume processing.
User confirms FBR reconnection.
MCV DO
Starts Stabilization Delay Duration timer and MCV DO and Gas control
equipment module with output held at value recorded.
When timer has elapsed, resumes normal automated control of
equipment module setpoints.
FBR DO
Starts the FBR DO equipment module by ramping the recirculation
pump from the bioreactor DO controller minimum flow rate to the
recorded output. The time to ramp is set by the Recirculation Pump
Ramp Duration setpoint. When the Stabilization Delay Duration and
the Recirculation Pump Ramp Duration timer have elapsed, resumes
automated control of equipment module setpoints.
Temperature
Starts the MCV Temperature and FBR Temperature control equipment
modules with both of their outputs held at the recorded controller
values until the Stabilization Delay Duration has elapsed. When
timer has elapsed, resumes normal automated control of equipment
module setpoints.
pH
Reads output value recorded.
If recorded output is >50: Starts equipment module pH control with
its output held at 50% until Stabilization timer has elapsed. When
timer has elapsed resumes normal automated control of equipment
module setpoint.
If recorded output is ≤50: Starts equipment module pH control with
its output held at the value recorded until Stabilization timer has
elapsed. When timer has elapsed resumes normal automated control
of equipment module setpoint.
Prompts user that system has stabilized, and previous phase is
resumed.
User acknowledges Substrate Sampling Phase
completion prompt.
