Use the
F
,
C
and
L
buttons to get images from the first slice, center slice and last slice of the stack. The
Focus Position
updates accordingly. The
Offset
value then might be set to half of the Interval
depending on the interval value and the number of slices, as with a lower number of slices or a large
interval the real center position of the stack might be just between two slices.
When the
Continuous Drive
checkbox is ticked the Z-Stack is performed using a continuous movement
of the z-drive. The stack size is retained. The z drive will move the distance of the optical slice thickness
during one exposure time that is set in the
Channels tool tab
. However, the overlap of two adjacent
frames resulting from the interval distance is kept in this mode.
When the
Continuous Drive
is deactivated, each image is taken after the z-drive has come to a stop.
After the frame is imaged interval distance is travelled before stopping to take the next image frame of
the Z-Stack.
Using the Continuous Drive will therefore speed up acquisition of a Z-Stack immensely.

Multidimensional Acquisition –
Time Series
When the
Time Series
tool tab is expanded, the
Time Series
control panel becomes available
(Fig. 96). In the
Time Series
tool the parameters
for the sequential acquisition of image frames as
time series are defined.
When time series are acquired with the
Lightsheet Z.1 a master file is created, with the
following appearance: Name.czi (e.g.
FishEmbryogenesis.czi). For each time point, a
separate file is additionally formed in the same
folder with the nomenclature:
Name(time point).czi
(e.g. FishEmbryogensis(221).czi).
To set up a time series acquisition, first you define the
Duration
it should acquire images by using the
slider or type into the text field, or use the arrow buttons. Next to these is a selection box in which you
can choose the dimensions corresponding to the set
Duration
; either cycle, msec (milliseconds), sec
(seconds), min (minutes), h (hours), d (days).
A maximum value of 100,000 can be set. The amount of data that can be handled by the system is
limited. If the maximum amount is exceeded, a warning will appear. In this case, it is necessary to either
change the destination for data storage and/or reduce the amount of cycles. The limit also depends on
the imaging setup and the acquisition mode.

checkbox is activated the system will continue taking images until the
available storage is filled.
Interval
defines a potential time interval, set in milliseconds (ms), seconds (sec) or minutes (min) by the
Interval
drop down menu, to separate the beginning of two cycles. A time shorter than the imaging
time for a frame is not effective. The interval time can be changed by using the
Interval
slider or
Interval
input box, also while imaging.
The
As fast as possible
checkbox sets the
Interval
slider to 0,0.
