Priming the Hydraulic System
Checking Prime
The following procedure is used to verify that there is no air in the hydraulic
system. Air in the hydraulic system can cause inaccurate supernatant-out volumes
and Excess Pressure alarms.
Note: When
using the priming
cushion, always use
the priming disk. Without
the priming disk, friction will
make it difficult to remove
the centrifuge bowl cover.

If the priming cushion and priming disk are not installed, do the following,
otherwise proceed to step 2.
a Open the centrifuge cover interlock latch (if applicable), open the sliding
covers, and remove the centrifuge bowl cover and alignment blocks.
b Place the priming cushion and priming disk in the centrifuge bowl.
Ensure the priming disk is on top of the cushion (Figure 6-22 on
page 6-36).
c Replace the alignment blocks and the centrifuge bowl cover.
d Close the sliding covers and the centrifuge cover interlock latch (if
applicable). Verify that the READY light is illuminated.

Set the main control panel and the secondary control panel to the following
settings:
• CENTRIFUGE SPEED = 3000 rpm
• SUPER OUT RATE = 450 mL/min
• SUPER OUT VOLUME = 600 mL
• AUTO/MANUAL = AUTO or MANUAL
• PUMP RESTORE RATE = (adjust to maximum setting)

Press the START SPIN button and allow the centrifuge to spin for at least
15 seconds.

Press the SUPER OUT button.
Note: Gambro
BCT suggests
checking prime if
your COBE 2991 has not
been used for 48 hours.

Verify that the EXCESS PRESSURE light illuminates within 15 seconds after
you press the SUPER OUT button, and then do the following:
• If the EXCESS PRESSURE light illuminates within 15 seconds, the
COBE 2991 is primed and ready for use.
-or-
• If the EXCESS PRESSURE light does not illuminate, the COBE 2991 is
not primed. Proceed to “Priming” on page 6-36.
Caution:
Do not operate
the COBE 2991
with the centrifuge bowl
empty, with an empty cell
processing bag, or without
the priming cushion. This
will cause the flexible
diaphragm to rupture.
