When air and starting cell product reach the cell product bag, press the
BLOOD IN button again. Additional starting cell product enters the cell
processing bag.

When no more fluid will flow into the cell processing bag or the fluid level
(Figure 6-15 on page 6-17) is just above the rotating seal, press the STOP
RESET button.
The capacity of the cell processing bag is approximately 630 mL. If the volume
of the starting cell product is more than 630 mL, not all of the starting cell
product will flow into the cell processing bag during the Blood-In/Air-Out
sequence. Additional starting cell product may be added during subsequent
Agitate/Wash-In operations by programming valve V1 to open instead of
valves V2 or V3. Add additional wash cycles as needed.

If desired, rinse the cell product bag (to avoid cell loss) by doing the following:
a Place a clamp on the clear tubing between the junction manifold and the
RCD.
b Place the cell product bag on the tabletop (Figure 6-17 on page 6-19).
c Press the PREDILUTE button and allow the desired volume of
processing/wash solution to fill the cell product bag.
d Press the STOP RESET button and manually agitate the cell product
bag.
e Hang the cell product bag on the left hanger bar.
f Press the BLOOD IN button, let the cell product bag empty, then press
the STOP RESET button.

Reset the CENTRIFUGE SPEED control to the appropriate setting,
according to the protocol.

Proceed to “Processing” on page 6-26.
Predilution Procedure Using the Blue-Striped Tubing
Note: This
procedure applies
only to protocols
where the starting cell
product is connected to the
red-striped tubing.
Use the following procedure to predilute the starting cell product with
processing/wash solution connected to the blue-striped tubing.

Ensure that the blue-striped tubing is not loaded into the collect valve (if
applicable). To open the collect valve, press the TUBE LOAD button, set the
VALVE SELECTOR to V1, and place the COLLECT/SOV button (toggle
switch) in the COLLECT position.

Clamp the clear tubing between the RCD and the rotating seal
(Figure 6-16 on page 6-18).

Place the cell product bag on the tabletop (Figure 6-17 on page 6-19).

Close the slide clamp on the blue-striped tubing. Remove the cap from the
spike and insert it into the processing/wash solution container. Hang the
container on the left hanger bar.
Caution:
Verify that fluid is
visible above the
rotating seal when the
centrifuge is spinning to
ensure adequate seal
lubrication. Inadequate
rotating seal lubrication
can lead to failure of the
rotating seal and potential
contamination or loss of
cell product.
