Processing – Experimental PSF

This function allows determining the point-spread
function (PSF) of sub-resolution beads and using
this experimentally determined PSF as input for
Structured Illumination processing.
Select the
Experimental PSF
tab to select the
Experimental PSF
Processing tool (Fig. 330).
Activate the image (a 3D stack of sub resolution
beads) you want to process in the image container
and press the
Select
button for
Input Image
to
load the image. Then expand the
Method
Parameters
tool window to have access to setting
parameters (Fig. 331).
You can load a 3D stack of a widefield
image or a non-processed raw data SIM
image. LSM image formats are not accepted
to determine the
Experimental PSF
.
You have the possibility to automaticaly compute
the Experimental PSF by choosing
Automatic
from
Mode
drop down menu (Fig. 332).
If you want to process manually and set quality
parameters, select
Adjust Parameters
If you want to edit selected beads you can do so
when selecting
Adjust ROIs
. Here, all selected
beads (white circels) will be displayed in the input
image as well. You can select a circle by clicking on
it and delete it by pressing the Delete button of the
keyboard. Alternatively, you can go from the
Preview view tab to the 2D view tab were all
selected beads are circled and listed and delete
circles from there. The beads of deleted circles will
then not be taken into account for the
Experimental PSF calculation. On the other hand
you can circle a bead in the 2D view not selected
before and add it in this way for claculation of the
Experimental PSF. Circles should be matched in
radius to the preselected circles.
If you want to define suitable beads by yourself,
you should select
Manual ROIs
. Here, no beads
are selected which then can be done in the 2D
view tab by choosing the circle graphic element for
an overlay.
