Pulse Measurements
The electronics system measures pulses by three characteristics: Height, Area,
and Width, as shown in Figure 15.

Pulse Height is the maximum digitized intensity measured for the pulse.
•
Pulse Area is an integration of the digitized measures of the pulse over time.
•
Pulse Width is a time period within which the pulse measurement is above the
threshold.
Electronics system calculates the Height and Area for FSC, SSC, and every
selected fluorescence channel. Width is based on the FSC signal. One effective
event has one Width parameter.
Pulse Digitization
The electronics system digitizes the pulse produced by the photodetector. The
digitized data is stored in memory and further processed to calculate the pulse
Height, Area, and Width. A high-accuracy high-speed 24-bit analog-to-digital
convertor is used to process the pulse in a speed of 80 million times per second.
Each pulse signal is divided into 16,777,216 (2
²⁴
) levels (Figure 16). The Height,
Area, and Width parameters of all available channels for each processed cell or
particle are then transferred to the NovoCyte Workstation for further display and
processing by the NovoExpress software.
