4.6 Quarterly maintenance
Condition the Pins
Condition the pins with 0.1 M NaOH on a quarterly basis (or as directed by Agena
Bioscience Customer Support). This is in addition to the daily pin cleaning. On the day
that pins are conditioned with NaOH, you should still perform pin cleaning with ethanol.
Perform the quarterly conditioning first and then the daily cleaning.

You will be handling NaOH in this procedure. For safe handling procedures, refer to
the Material Safety Data Sheet (MSDS) provided by your NaOH supplier. At a minimum: 1) wear
protective clothing (laboratory gloves, eye protection, and laboratory coat), 2) take extreme
caution to avoid eye or skin contact, 3) have adequate ventilation of your laboratory, and 4) have
appropriate spill kits readily accessible. If skin or eye contact occurs, flush with large amounts of
water for at least 15 minutes.
To condition the pins:
1.
Using a multi-channel pipette, add 30
L of 0.1 M NaOH to wells A1 through D6 (24 wells
total) of an empty 384-well microtiter plate
NOTE
will work for 24, 6, or 1-pin array formats. For the 6-pin format, only
wells A1, A3, A5, C1, C3, and C5 are used. For the 1-pin format, only well C5 is used.
NOTE
For a 6-pin instrument (or 24-pin instrument adapted to 6-pin format) fill wells A1-B3 (6 wells total)
of a 96-well microtiter plate with 120
L of 0.1 M NaOH. See

Tap
PARK
on any screen (located at the upper right screen). Parking the pin array
assembly allows easier access to the processing deck.
3.
Place the first microtiter plate on the left plate flattener or holder (depending on the
instrument version):

Fill each of these
wells with 30
L
of 0.1 M NaOH
This plate setup will work for 24, 6,
or 1-pin array formats.
Only wells A1, A3, A5, C1, C3, and
C5 are used for the 6-pin format.
Only well C5 is used for the 1-pin
format.
