Starting your run with
cells
1. Once your vessel/bioreactor is filled
with cell free media, the vessel
adaptor must be attached to the
vessel.
N.B.
The
OPTURA
SPY
must only be
used on optically clear vessels with
a wall thickness <4.3 mm
2. Flex the wings on the adaptor
before use using your thumbs to
gently work these back and forth.
3. Peel off the sticker from the back
of the adaptor and place firmly
onto your vessel in an area which
has 2.5 cm of free, unimpeded
space inside the vessel. Apply
pressure for 10 seconds.
4. Insert the OPTURASPY sensor
by removing the dust cap and
inserting the sensor into the
adaptor until you hear a click, the
sensor is keyed to be inserted in
one orientation.
5. Open the
OPTURA
VIEW
software as
described above and connect your
OPTURASPY.
6. In OPTURAVIEW, select the tab
<Live Readings>
, click
<Start
Recording…>
and enter your
own unique run ID, confirm the
logging interval, serial number and
whether the system checks have
been performed. Select
<OK>
to
start recording.
7. Your OPTURA system is now
recording.
8. When the readings are stable in
cell free media click
<Add Event>
and select
<Zero>
from the
drop down and then
<OK>
. This
eliminates background readings
from your measurements.
N.B.
All process conditions should be
set and stable when performing
the zero. Zeroing should be the
last action performed prior to
inoculation.
9. Monitor live trend for a couple
of minutes to verify that zeroed
reflectance readings are stable and
re-zero if necessary.
10. Inoculate your bioreactor.
11. The software should now trace the
growth profile of your process.
12. Click <Stop Recording> only when
you have completed your run.

If you are calibrating reflectance
readings to offline readings,
follow the Biomass Calibration
instructions in the manual
