Pressing the Optimal button
sets
Interval
to a value of half the optical slice thickness. Two adjacent
slices have therefore always a 50% overlap. The value in
Slices
is adjusted accordingly to keep the
Range
within the limits defined by
Set First
and
Set Last
. The optimal interval is displayed within the
button in
µm
.
Within the
Keep
display field (only available in
Show all
mode) you have the option to switch between
Keep
Interval
and
Keep Slice
. If
Keep Interval
is checked, the number of slices is adjusted to a new
Range
defined by pressing
Set First
or
Set Last
. Likewise, when
Keep Slice
is checked, the value for the
Interval
between the slices is adjusted to a new Range defined by pressing
Set First
or
Set Last
.
The
Position (µm)
input box indicates the current position of the focus. The number can be edited by
using the arrow keys. The focus drive will automatically move to the new position.
When the
Continuous Drive
checkbox is ticked the Z-Stack is performed using a continuous movement
of the z-drive. The stack size is retained. The z drive will move the distance of the optical slice thickness
during one exposure time that is set in the
Channels tool tab
. However, the overlap of two adjacent
frames resulting from the interval distance is kept in this mode.
When the
Continuous Drive
is deactivated, each image is taken after the z-drive has come to a stop.
After the frame is imaged, the interval distance is travelled before stopping to take the next image frame
of the Z-Stack.
Using the Continuous Drive will therefore speed up the acquisition of a Z-Stack immensely.
The
Z-Stack Navigation
field displays a graphical representation of the Z-Stack. The
white frames
represent the number and localization of the slices. The
blue frame
indicates
the current focus position,
also during image acquisition. By dragging the
blue frame
with the cursor, the current focus position
changes accordingly. The
ruler
immediate below the graphical representation indicates the dimensions
(range) of the Z-Stack.
The
F
,
C
and
L
buttons indicate the positions of the first slice, center slice, and last slice of the stack,
respectively. If you press one of the buttons, an image will be acquired at the respective focus position
It is not relevant which of the chosen slices is assigned first and last position. The system will always move
the sample away from the detection optics when acquiring the stack.
