Calibration of the Stage
In order to calibrate the stage, press the
Calibration
Home XYZ
button of the
Stage LLS
tab in the right
tool area (Fig. 49). This is an optional step but will
zero the stage in X, Y and Z and displayed positions
are then absolute numbers.
We recommend removing any sample from
the stage during homing as the stage will
travel long distances and the Immersion might
brake away. Also, the carrier might shift in
position.
After homing the system will have absolute
positions. This might be helpful especially for
z-positioning using the same Carrier when cells
are sparse.

Priming immersions
There are two possibilities to prime immersion,
manual with the syringe and by software via the
pump. For manual priming push the piston of the
filled syringe. Inspect the immersion media emerging
on top of the meniscus lens. If air bubbles are visible,
suck off immersion with a soft tissue paper and
repeat, until immersion is air bubble free. For priming
via the pump press the
Prime Pump
button of the
Auto Immersion
tab of the right tool area (Fig. 50).
As the pump can be too weak to press out any
air bubbles within the tubing, we recommend
using the syringe for immersion priming
For short time experiments without heating it
is enough to apply water immersion with a
pipette from the top.
