Acquire and Analyze a Chemical Image
Create a chemical image to identify components of your sample or to gain understanding of
the sample’s structure and composition.
 To acquire and analyze image data
1. Load the sample onto the microscope stage.
2. Configure sample illumination .
3. Click the Video button in the software to select video mode.
4. Bring the sample into focus and collect a mosaic or a terrain mosaic.
5. Use Live spectrum mode to acquire spectra at several locations of the sample in order to
optimize the Laser power , Exposure time and Number of scans settings.

Feature Description
Photobleach time Time for each sample point to be exposed to the laser for
photobleaching before collection begins. Adding photobleach
time may reduce sample fluorescence but will also increase scan
time.
EM gain Controls the electron multiplying gain of the detector camera.
EM gain increases the signal to overcome the readout noise, but it
also increases other sources of noise. The optimal setting for this is
highly dependent on the experiment. In general, it is
recommended to run with EM gain on for most operation but to
turn it off when collecting with a longer exposure time.
Laser polarization
(requires polarization
option) Sets the polarization for the laser light incident on the sample
Analyzer polarization
(requires polarization
option) Sets the position for the polarization analyzer controlling the
Raman emission. This option is only available when Laser
polarization is set to "Horizontal(X)" or "Vertical(Y)" to run a
polarized experiment.
Analyzer angle
(requires polarization
option) Sets an arbitrary angle (0–180 degrees) for the polarization
analyzer controlling the Raman emission. This option is only
available when Analyzer angle is set to "Select Angle."
