5.4.1 Assembly of the PyroMark Q96 Vacuum
Workstation
Assemble the PyroMark Q96 Vacuum Workstation as
follows:
1.
Place the PyroMark Q96 Vacuum Workstation on a flat
surface.
2. Connect vacuum tubing to the straight blue connector on
the cap of the liquid waste container. Attach the other
end of this tubing to the vacuum pump.
3. Connect a second piece of vacuum tubing to the angled
blue connector on the cap of the liquid waste container.
Attach the other end to the connector on the back of the
PyroMark Q96 Vacuum Workstation.
4. With a third piece of tubing, attach the Vacuum Prep
Tool to the vacuum tubing connector on the top of the
PyroMark Q96 Vacuum Workstation (see image below).
5. Test the PyroMark Q96 Vacuum Workstation by switching
on the vacuum pump and opening the switch on the
front of the workstation. If not vacuum is created, turn off
the vacuum pump and open the cap of the liquid waste
container to release any built-up air pressure.
Note : The liquid waste container must be able to
withstand absolute vacuum.
5.4.2 DNA amplification
Amplify the DNA to be analyzed by PCR using one of the
primers biotinylated. See Appendix B for details on assay
design and validation.
