10.
Select the Setup Microscope tab.
You are prompted to start the microscope initialization and aperture calibration
procedure.
11.
Make sure that the lower cassegrain is fitted, that if the ATR objective is fitted it is
retracted, that the stage is centered and that there is no sample on the stage, and then
click
OK
.
The microscope initializes and runs the aperture calibration procedure.
NOTE:
If you do not want to initialize your instrument at this time, you can click
Cancel
.
You will not be able to use any of the microscope controls on the Setup Microscope
tab until you have initialized the microscope. To initialize the microscope later, click
Initialize
on the Setup Microscope tab.
Focusing the Microscope
Microscope sample positioning and focus controls
The focus of the microscope is changed by moving the sample stage up or down. While
focusing, you can view the image on the Setup Microscope tab.
Turn the coarse or fine focus controls (Figure 13) clockwise to move the stage up and
counter-clockwise to move the stage down.
The default position for the lower cassegrain gives optimal illumination for thin samples. For
optimal performance it may be necessary to refocus the lower cassegrain to compensate for
the sample’s refractive index.
