7.2. Decalcification guidelines
Under mentioned indicative time-range are for generic bone samples. Temperatures and times have to be
validated according to bone type and to the quantity of present calcium
Milestone recommends the checking of the bone samples during a decalcification program at half of the
above time, so that users can better define optimum protocols for their samples.
MoL-DECALCIFIER: recommended solution for optimal morphology, IHC and molecular biology.
Heated decalcification with both MoL-DECALCIFIER and Formic acid leads to satisfactory morphology, IHC
and molecular biology but it’s of paramount importance not to over-decal tissue, hence the checking of the
bones’ status must be consistently performed.
When users decide to set their own protocols, temperatures above 50°C should be avoided.
