Solvent mixtures
When creating methods for solvent mixtures, linked runs can be used to optimise the
evaporation process; volatile components can be evaporated and then removed from the
condenser before a high vacuum stage completes the non volatile part of the mixture.
Note:
Runs set up using auto program software, will consist of a single stage optimised for
the higher boiling point solvent. While the feature offers a quick set-up, it is invariably more
efficient to create methods by linking multiple runs together.
The following example outlines a general method that could be used to evaporate a mixture
of DMSO and DCM. Specific method settings would depend on variables such as volume,
mixture ratio and the type sample holder used.
1.
Select
Variable Dri-Pure
and set a ramp from 750 mbar to 70 mbar in 55 minutes. This
helps to prevent bumping.
2.
Evaporate the DCM at 65 mbar; it will collect in the condenser.
3.
Select
Auto-Defrost and Drain
. This drains the organic solvent from the condenser at
the end of the stage, allowing high vacuum levels to be achieved later when removing
the high boiling point solvent.
4.
Select
Full Vacuum
to remove the high boiling point solvent, end the method using
Heat-Off Option

Heat Flow
.
5.
Dry any stubborn leftover solvent. Select
Full Vacuum
and let it run for one hour.
6.
Enable
Auto-Defrost and Drain
for the final drying stage, this initiates a full condenser
defrost and drain at the end of the process.
