8 Concentration analysis
8.3 Evaluating calibration-free measurements

Biacore T100 Software Handbook BR-1006-48 Edition AE

The fitting procedure is normally fast. If fitting appears to hang for a particular
sample series, you can click

Accept

to accept the current fitting results for that

series or

Abort current

to abandon the series. Click

Abort remaining

if you want

to abandon all remaining series in the evaluation. The fit status is recorded in the
table of results.

Click

Expand all cycles

in the finished result display to display the details of the

sample series in the cycle list.

The results are reported in terms of

Measured Conc

, which is the value

calculated from the fitting, and

Calc Conc

which is obtained by multiplying the

Measured Conc

by the dilution factor to give the concentration in the original

sample. You can change the units for the reported concentration in the header
for the

Measured Conc

column.

Note:

The diffusion coefficient

listed in the evaluation results is the value at

the analysis temperature, calculated automatically in the software from
the value at 20°C provided when the assay is run.

8.3.4

Interpreting the results

Consider the following aspects of calibration-free concentration analysis when
interpreting the results:

•

Samples where the QC ratio of the i is less than about 0.13 should be
discarded (see Section 8.3.1). Results for samples where the initial binding
rate at 5 µl/min is outside the range 0.3–15 RU/s should be treated with
caution.
