5.4.5 Annealing of sequencing primer to samples

The thermoplate and the heating block (for annealing) can
reach temperatures of up to 80°C (176°F). Avoid touching
them when they are hot.
1.
Place the PyroMark Q96 Plate Low in a prewarmed
PyroMark Q96 Sample Prep Thermoplate Low, and heat
the samples on a heating block at 80°C for 2 min.
2. Remove the plate from the thermoplate and allow the
samples to cool to room temperature (15–25°C) for at
least 5 min. The plate can now be processed in the
PyroMark Q96 ID.
5.4.6 Reagent preparation
1. Prepare reagents according to instructions included with
the PyroMark Gold Q96 Reagents.
2. Open a reagent cartridge pack and take out a new
cartridge. The cartridge has to be handled with care in
order to maintain accuracy.
3. Remove the plastic needle cover from the underside of
the reagent cartridge. Save the cover for use when
disposing of the reagent cartridge. Do not use the plastic
needle cover between runs, since applying and removing
the cover several times may damage the needles.
Note : If re-using a reagent cartridge, ensure that it is
cleaned appropriately, see Section 7.6. If handled
correctly, a reagent cartridge can be used for up to
20 runs.
Note : It is important that the reagents are pipetted into
the correct reagent cartridge compartments.
4. Place the reagent cartridge with the label facing towards
you. Pipet the recommended volume of substrate solution
into the
S compartment of the reagent cartridge.
