Verifying Shading Correction Files-Legacy
Note: This procedure is required if you select the Legacy Correction check box for one
or more wavelengths in an acquisition protocol.
If you select Legacy Correction as the Shading Correction method for an acquisition
wavelength, you need the shading correction image files for each magnification and
illumination setting. You then use these files to generate the files again whenever you replace
an objective, a filter set, or the light guide. See
Updating Legacy Shading Correction Settings
on page 233 .
To verify that the shading correction image files are available for the Legacy Correction
setting:
1. In the MetaXpress software, do one of the following to display the Plate Acquisition Setup
dialog:
From the simplified menu, click Screening > Acquisition Setup.
From the standard menu, click Screening > Plate Acquisition Setup.
2. Select the Configure tab.
3. On the Configure tab, select the Acquisition tab.
4. Click Directory for Stored Correction Images to display the Browse for Folder dialog. The
default location for stored correction images is C:\Shading Images.
To ensure that the file names are formatted correctly:
1. In Windows, open File Explorer and navigate to the C:\Shading Images folder (or a defined
location).
2. Locate the shading correction files.
3. Ensure that a file exists for each magnification and illumination setting.
Shading correction images use the following naming format:
For Widefield:
shading_ <magnification name> _widefield_ <illumination
setting> .tif
For example:
shading_4x Plan Apo_widefield_DAPI.tif
For Confocal:
shading_ <magnification name> _confocal<disk size>um<slit or
pinhole>_ <illumination setting> .tif
For example:
shading_20x_confocal 60um pinhole_FITC.tif
