Capturing a multi-color z-stack

Select
Measurement Mode > Multi Color Acquisition
in the
Measurement Wizard
window.

Select
xyz-Table Z
and
UltraII Filter
in the
Devices
list.

Click the
Autosave
button at the end of the row under
AS
. It will turn green.

Select the correct excitation and emission wavelengths for your sample under
Filter for Measurement
in
the
Settings 1
window. The selected checkboxes are used.

Move the slider under
Laser Transmission Control
while the filter is highlighted to set the laser power for
each individual filter.
NOTICE!
Higher laser power will bleach the sample.

Select the shortest emission wavelength, and create a sharp picture.

Correct the different focus lengths for different emission wavelengths using the
Chromatic Correction
button, starting with the next longer wavelength.

If all emission wavelengths have been corrected, click
Save Settings
in the toolbar.

To start the measurement, proceed as described under
Capturing an image on the previous page
