Managing Plate Models
Overview
For different experiments, researchers use different kinds of plates with different shapes, depths and
characteristics: v-bottom vs. round-bottom vs. flat bottom; shallow vs. deep; circular vs. square. This article discusses
how to set up and maintain a “library” of plate models that can be used when defining an experiment protocol.
Understanding Plate Model Calibration
The plate model controls the sampling depth. Each plate model has a
depth offset
which determines the probe
position during sampling. This offset is a measure of how far above the deck the probe should go. Sampling Height is
calculated as follows: Deck Depth Offset = Sampling Height.
With the Gen 1 probe assembly, a single depth offset is calibrated for each plate model. With the Gen 2 probe, the
Plate Model is mapped generating a series of Depth Offsets which provides the more precise sampling required
for 1536 well formats and also which accounts for possible variations in the alignment of the sample arm and the
shaker deck.
In either case, proper plate model calibration is required for accurate sampling.
NOTE
Proper plate model calibration is essential. If the probe hits the bottom of the
well during sampling, it is possible that no sample will be aspirated, probe tip
clogging, or that probe damage could occur.
