Start Transform1 File Creation
WHY DO THIS?
The goal here is to remove the distortion caused by the fiber optic taper. This is accomplished by
capturing a raw image of the bebe grid which is a known standard (equally spaced bebes). Using
the Transgen program, we can specify the distance between pixels on the detector - therefore,
when we unwarp an acquired image, the corresponding pixels will be properly represented in
MicroView. The end result means that when we unwarp the acquired bebe grid, it will be an
accurate representation of the actual (physical) grid.
To start the Transform1 file creation:
1.
Correct the grid image using the
Calibration Tools
program as follows:

click on the
Correct
tab
-
in the VFF to Correct field, find the
newly captured image
-
use the
Browse
buttons to find the
bright
and
dark
files created earlier
-
click the
Correct
button (the
corrected file is named
corrected.vff
and it is stored in the
same folder in which the captured
imaged was stored)
2.
Double-click on
corrected.vff
in
Windows Explorer
to load the image into
MicroView
and
confirm that the grid has been corrected (image looks cleaner with less noise in between the
bebes).
3.
Double-click on the W and L parameters in the lower right of the screen. Change the Update
mode to
Delayed updates
. This only needs to be changed once.
