•
Unpack and empty the column. Run a pressure drop test over a range of flow rates and
compare results to pressure/flow curves in this manual.
•
Ensure all connections to/from the column are correctly connected and correct size
pipework and ladish seals have been used.
•
Ensure any valves fitted to the column are correctly functioning.
•
Rectify any issues that are observed and repeat the pressure/flow test.
If process feed streams are either highly viscous, contain particulates or may precipitate it is
possible that the bed supports have become blocked. This can also occur if the chromatographic
media contains fines.
If this is suspected replace the bed supports and repeat the pressure flow test.
Media Leakage
There can be several causes of media leakage:
1.
Fixed flow cell is incorrectly positioned or not tightened correctly.
2.
Damage to the fixed bed support or integral seal.
3.
Damage to the top bed support or integral seal.
4.
Damage to the adjuster seal or misalignment of the seal causing failure of the seal
setting mechanism.
5.
Check that the particle size of media is suitable for the pore size of bed support.
6.
Remove all media from the column and inspect the column.
7.
Replace components as required.
