Creating an
AutoDelta Step
An AutoDelta step allows you to increase or decrease the temperature and/or change
the duration of the cycle during a cycling stage.
One use for an AutoDelta step is for touchdown PCR, a method in which the earliest
cycles have a high annealing temperature, followed by cycles where the annealing
temperature decreases slightly for each cycle. Touchdown PCR increases the
specificity of the PCR products. The high initial temperature favors more specific
annealing between the primer and the template. Because these fragments are further
amplified during subsequent cycles at lower temperatures, they outcompete the
nonspecific sequences to which the primers bind at lower temperatures.

AutoDelta steps are allowed only in cycling stages.
