Note:
Before starting the calibration, the instrument may pause (up to
10 minutes) to allow the heated cover to reach the required temperature.

When the run is complete and the software displays the Analysis screen, select
each filter from the Filter Set drop-down list, then confirm that the
corresponding ROI Image displays a green circle around each well area.

After you inspect all ROI images, confirm the status of the calibration, where
passed indicates that the run produced viable calibration data, and failed indicates
that the run did not produce data or that the data it collected is unusable.

Troubleshoot the failed calibration (see “Troubleshooting“).
10.
Unload the calibration plate or array card:
a.
After the calibration, tap on the touchscreen to eject the plate or array
card.
Note:
If the instrument does not eject the plate or array card, remove it as
explained in “Troubleshooting“.
b.
Remove the ROI calibration plate or array card from the instrument tray and
store it in the dark at room temperature until you are ready to perform the
uniformity calibration.
WARNING!
PHYSICAL INJURY HAZARD. During instrument
operation, the plate or array card temperature can reach 100°C. Allow
the consumable to cool to room temperature before removing.
c.
Tap on the instrument touchscreen to close the instrument tray.
11.
In the ROI Calibration screen, click Finish to complete the calibration, then click
Yes when prompted to save the results.
Note:
If the QuantStudio
™
Real-Time PCR Software displays the warning
Saving ROI calibration will invalidate other existing calibrations except HRM
Calibrations, click OK to proceed.
Note:
HRM (High Resolution Melt) is a separately licensed module. If your
instrument requires this module, contact Thermo Fisher Scientific.
12.
(Optional) Click Print Report in the upper right corner of the screen to print a
summary of the calibration results for your records.
Background calibration
The software uses the background calibration data during subsequent experiments to
remove background fluorescence from the experiment data.
